Beskrivelse

The main objective is to study protein-detergent interactions, both in simple mixtures and under more application-relevant conditions. We will identify which detergents interact most favourably with lipases under application-relevant conditions, in order to obtain the largest effect at the lowest possible detergent concentrations. We intend to transfer the experience within the AAU group to model systems chosen in collaboration with participating members of the consortium. A broad array of proteins will be selected, based both on basic and application-oriented criteria. As far as possible we will work with simple systems (one protein, 1-3 different detergents); this will be expanded with time to include application-relevant conditions. We will hold regular status meetings involving the participating members, combined with a steady information flow (bearing confidentiality issues in mind).     A thorough analysis of protein-detergent interactions will be carried out, in which binding and unfolding will be studied with all possible techniques. In this way we hope to obtain a better understanding of the molecular basis of these interactions.
